Hi,
I have a custom icon I am using for an app in quick start and its background
is presently white, so it is showing as a square. I would like to make it
transparent for this purpose. which color do I need to make it (In RGB)

You don't set a color, you set the background to be transparent.

If your tool can't do this try

When I reduce the icon to 256 colors it ruins it, so I will leave it as is.
